Nicotinamide mononucleotide (NMN, β-NMN) is a direct precursor to NAD+, the coenzyme that drops off as we age. Most research has used doses in the 250–500 mg/day range. Here the calculator scales by body weight: dose_mg = weight_kg × factor_mg_per_kg. At the default 4 mg/kg, a 70 kg adult comes out near 280 mg/day, right inside that most-studied band.

NMN got its moment in the spotlight after David Sinclair’s lab at Harvard Medical School tied NAD+ restoration to mitochondrial function, sirtuin activation and biomarkers of biological aging. A couple of regulatory points to keep in mind: in 2022 the U.S. FDA pulled NMN out of the supplement category, citing its status as an investigational drug, and Brazil’s ANVISA has never registered NMN as a food supplement. Long-term human safety data are still thin. Even so, the global NMN market hit roughly US$400 million in 2024.

FAQ

Is NMN approved by regulators? No. The FDA reclassified it as an investigational drug in 2022, and ANVISA hasn't registered it as a supplement. In several countries it sells in a regulatory gray zone.

What’s the difference between NMN and NR? Both feed into NAD+. NR (nicotinamide riboside) is one phosphate group smaller and sells in the U.S. as a registered supplement, Niagen. NMN sits one step closer to NAD+ along the salvage pathway.

Are the longevity claims proven? Animal studies (mice) are promising, but robust long-term human RCTs are still scarce. Treat NMN as experimental and consult a clinician.
